Ketotifen belongs to the group of stabilizers of mast cell membranes. It inhibits the release of histamine and other mediators (including leukotrienes, cytokines) from mast cells, prevents eosinophilic infiltration in places of allergic inflammation. It has blocking effect on (H1) histamine receptors, inhibits phosphodiesterase, blocks calcium channels of cells, is a functional antagonist of leukotrienes.
Ketotifen is an effective oral agent for preventing bronchial asthma. It has a pronounced anti-anaphylactic and antiallergic effect. When used alone, it does not stop asthma attacks, but leads to a decrease in the number, duration and intensity of these attacks. In some cases there is a complete disappearance of asthma attacks. The drug favorably influences the excretion, reduces eosinophilia, reduces the need for the use of bronchodilators, corticosteroids, etc.
Antiallergic agent. The mechanism of action is associated with the stabilization of mast cell membranes and a decrease in the release of histamine, leukotrienes and other biologically active substances from them. Suppresses the accumulation of eosinophils in the airways caused by platelet activating factor. Prevents attacks of bronchial asthma, some other manifestations of allergic reactions of an immediate type. Blocks histamine H1 receptors.
After oral administration, it is almost completely absorbed from the digestive tract. Simultaneous eating does not affect the degree of absorption of ketotifen. It is metabolized by 50% during the "first passage" through the liver. Cmax in plasma is reached within 2-4 hours. Binding to plasma proteins is 75%. Ketotifen is excreted from the body in two phases. The half-life in the initial phase is 3-5 hours, in the final phase - 21 hours. It is excreted by the kidneys, 60-70% in the form of metabolites, 1% - unchanged.
Take inside. Adults - 1 mg 2 times / day (morning and evening) with meals. If necessary, the daily dose can be increased to 4 mg.
Maximum daily doses: for adults - 4 mg.
For children aged 3 years and older - 1 mg 2 times / day; at the age of 6 months to 3 years - 500 mcg 2 times / day.
Ketotifen can enhance the effects of sedatives, hypnotics, antihistamines and ethanol.
When taking ketotifen simultaneously with oral hypoglycemic drugs, a reversible decrease in platelet count was noted.
- From the side of the central nervous system: drowsiness, slight dizziness, slowing of mental reactions, usually disappearing a few days after the start of treatment.
- From the digestive system: increased appetite; rarely - dyspeptic symptoms, dry mouth.

Pregnancy and lactation
During pregnancy, especially in the first trimester, use is possible only if the potential benefit to the mother outweighs the potential risk to the fetus.
Ketotifen passes into breast milk, so if necessary, use during lactation should decide whether to stop breastfeeding.
The therapeutic effect of ketotifen develops slowly, within 1-2 months.
The ongoing anti-asthma therapy should be continued for at least 2 weeks after the start of ketotifen administration.
With the simultaneous use of ketotifen and bronchodilators, the dose of the latter can sometimes be reduced.
